Kambosos made his professional boxing debut against Jayson Mac Gura of the Philippines. Kambosos, who was 19 at the time, won via technical knockout in the second round. He defeated world number nine Brandon Ogilvie to win his first championship, the WBA Oceania. In May 2017, he defeated Qamil Balla in a ten-round bout, with Kambosos winning by unanimous decision.

He won the WBA Oceania and IBF Pan Pacific titles in the lightweight division five months later, defeating Krai Sethaphon in the ninth round. Manny Pacquiao recruited him as his main sparring partner in June 2017 in preparation for his fight with Jeff Horn, and he has been Pacquiao’s main fighting partner for his subsequent fights.

In May 2018, he made his US debut, defeating Jose Forero in just 1 minute and 48 seconds. On January 19, the following year, he defeated Rey Perez on the undercard of Manny Pacquiao vs. Adrien Broner at the MGM Grand in Las Vegas. In an eight-round battle, he won by a majority vote.

On June 7, 2019, in the Galatsi Olympic Hall in his home Greece, Kambosos beat off Venezuelan 11-2 Ricard Pena in round six. In December of the same year, he won a split decision against Mickey Bey, the former IBF lightweight champion, on the undercard of Terence Crawford versus Egidijus Kavaliauskas.

Kambosos won a split decision over Lee Selby (former IBF featherweight champion) at the SSE Arena in London on October 31. With the victory, George became the mandatory challenger for Teófimo López’s IBF lightweight championship. The fight between George and Lopez was scheduled to take place at LoanDepot Park in Miami, Florida on June 19, 2021, but it had to be delayed as Lopez tested positive for COVID-19.

Nevertheless, they are rescheduled to fight on August 14, 2021.
